Pharmacodynamic studies show that tesamorelin reduces visceral adipose tissue more than subcutaneous adipose tissue in the studied population. This selectivity may relate to differences in blood flow and hormone sensitivity between fat depots. Effects on glucose metabolism and insulin sensitivity have been investigated, with some trials reporting modest changes and others showing stability. The precise relationship between growth hormone exposure, IGF-1 levels, and visceral fat loss remains an active area of analysis.
Tesamorelin binds to growth hormone-releasing hormone receptors on somatotroph cells in the anterior pituitary. Receptor activation increases intracellular cyclic AMP and promotes synthesis and secretion of growth hormone. Because the peptide mimics endogenous GHRH, it amplifies the normal pulsatile release of growth hormone rather than providing exogenous growth hormone directly. This upstream action distinguishes tesamorelin from recombinant growth hormone preparations and from growth hormone secretagogues that act at different receptors.

Lyophilized material is typically held under refrigeration between two and eight degrees Celsius, shielded from light and ambient moisture. Peptides of this size adsorb to glass and plastic, so working procedures often call for low-binding containers and as few transfers as possible. Absorbed water during weighing shifts the apparent mass of a sample, and controlling room humidity reduces that source of error. Once dissolved, solutions are kept cold and used within the interval printed on the accompanying label or certificate. Degradation accelerates markedly in dilute aqueous form.
Identity and purity are judged through a combination of chromatographic and mass spectrometric techniques. Reversed-phase high-performance liquid chromatography separates the intact peptide from truncated, oxidized, and deamidated variants, and the resulting peak-area percentages yield a purity figure. Electrospray ionization mass spectrometry confirms the expected molecular mass and can expose unanticipated modifications. Amino acid analysis and peptide mapping support sequence fidelity, while water content, pH, sterility, and bacterial endotoxin testing describe the physical and microbiological attributes of a finished lot.
Regulatory approval in the United States came in 2010, when the Food and Drug Administration cleared the peptide for the reduction of excess abdominal fat in adults with HIV infection and associated lipodystrophy. The decision rested mainly on two randomized phase 3 trials that enrolled roughly eight hundred patients and ran for twenty-six weeks. Participants receiving active drug showed substantially greater declines in visceral adipose tissue than those receiving placebo, while total body weight changed comparatively little. A reformulated presentation was later approved, and the product has remained a niche therapy rather than a general weight-loss agent.
Tesamorelin occupies a narrow position among agents that act on the growth hormone axis. Unlike growth hormone itself, which is given as replacement, it stimulates the pituitary to release the hormone in pulses, so the downstream increase in insulin-like growth factor 1 depends on intact somatotroph function. Other peptides in the same family include shorter GHRH fragments and synthetic secretagogues with different stability profiles. Several points remain unresolved, including whether the reduction in visceral fat translates into fewer cardiovascular events, what happens to metabolic markers after long-term use, and how the drug compares with lifestyle or surgical approaches.
Tesamorelin is a synthetic peptide that belongs to the growth hormone-releasing hormone family and contains the same forty-four amino acid sequence as endogenous GHRH, extended at the amino terminus by a trans-3-hexenoyl group. That small fatty acid modification protects the peptide from rapid cleavage by dipeptidyl peptidase-4, the enzyme that shortens the half-life of native GHRH to only a few minutes. Chemically the compound is produced by solid-phase peptide synthesis, purified by chromatography, and supplied as a sterile lyophilized powder for reconstitution.
Storage claims vary across suppliers, and published stability data for specific formulations are limited. Extrapolating from related peptides is common but not a substitute for direct measurement. For research use, documentation such as a certificate of analysis is often requested to confirm identity and purity. What constitutes an acceptable purity threshold depends on the intended application. Open questions remain about how temperature excursions during shipping affect long-term peptide integrity. Independent verification by an end user is not routinely reported.
Lyophilized tesamorelin is generally stored refrigerated at 2 to 8 degrees Celsius, protected from light and moisture. Peptides in this class are often kept frozen at minus 20 degrees Celsius for longer periods. Reconstituted solutions are typically used within a defined window because hydrolysis and oxidation proceed faster in liquid form. Container material and headspace also influence how long a preparation retains its expected profile. Specific stability figures depend on concentration and buffer composition.
Common analytical approaches include reversed-phase high-performance liquid chromatography for purity assessment and mass spectrometry for identity confirmation. Peptide mapping after enzymatic digestion can verify the expected sequence. Immunoassays may be used to measure the compound or its downstream markers, but they can cross-react with related peptides and require careful validation. Impurity profiles typically include truncated sequences, oxidized methionine residues, and residual solvents from synthesis. Each method reports a different property, so no single assay establishes overall quality.
Physicochemical behavior is dominated by the peptide backbone. The molecule is hydrophilic and carries a net positive charge near neutral pH, owing to several arginine and lysine residues. In solution it adopts a largely unstructured conformation, and aggregation is a known concern for peptide products of this size. Oxidation of methionine and deamidation of asparagine or glutamine residues are the principal chemical degradation routes. These liabilities shape how the material is formulated, handled, and analyzed, and they explain why lyophilized presentations are common in research settings.
Tesamorelin is a synthetic peptide analog of growth hormone-releasing hormone, built from 44 amino acids. Its sequence follows the natural human GHRH(1-44) backbone, with a trans-3-hexenoyl group attached to the N-terminal tyrosine. This modification blocks recognition by dipeptidyl peptidase IV, the enzyme that rapidly truncates the native hormone in circulation. The result is a molecule with a substantially longer plasma residence time than unmodified GHRH, which makes it practical for clinical and laboratory study.
Scientific literature indicates that some of the compounds of bismuth are less toxic to humans via ingestion than other heavy metals (lead, arsenic, antimony, etc.) presumably due to the comparatively low solubility of bismuth salts. Its biological half-life for whole-body retention is reported to be 5 days, but it can remain in the kidney for years in people treated with bismuth compounds. Bismuth poisoning can occur and has, according to some reports, been common in relatively recent times. As with lead, bismuth poisoning can result in the formation of a black deposit on the gingiva, known as a bismuth line. Poisoning may be treated with dimercaprol, but evidence for benefit is unclear. Bismuth's environmental impacts are not well known; it may be less likely to bioaccumulate than some other heavy metals, and this is an area of active research.
Religious leaders often turned to versions of exorcism to treat mental disorders often utilizing methods that many consider to be cruel or barbaric methods. Trepanning was one of these methods used throughout history. In the 6th century AD, Lin Xie carried out an early psychological experiment, in which he asked people to draw a square with one hand and at the same time draw a circle with the other (ostensibly to test people's vulnerability to distraction). It has been cited that this was an early psychiatric experiment. The Islamic Golden Age fostered early studies in Islamic psychology and psychiatry, with many scholars writing about mental disorders. The Persian physician Muhammad ibn Zakariya al-Razi, also known as "Rhazes", wrote texts about psychiatric conditions in the 9th century. As chief physician of a hospital in Baghdad, he was also the director of one of the first bimaristans in the world. The first bimaristan was founded in Baghdad in the 9th century, and several others of increasing complexity were created throughout the Arab world in the following centuries. Some of the bimaristans contained wards dedicated to the care of mentally ill patients. During the Middle Ages, Psychiatric hospitals and lunatic asylums were built and expanded throughout Europe. Specialist hospitals such as Bethlem Royal Hospital in London were built in medieval Europe from the 13th century to treat mental disorders, but were used only as custodial institutions and did not provide any type of treatment. It is the oldest extant psychiatric hospital in the world.
== Forms of topographic data == Terrain is commonly modelled either using vector (triangulated irregular network or TIN) or gridded (raster image) mathematical models. In the most applications in environmental sciences, the land surface is represented and modelled using gridded models. In civil engineering and entertainment businesses, the most representations of land surface employ some variant of TIN models. In geostatistics, the land surface is commonly modelled as a combination of the two signals – the smooth (spatially correlated) and the rough (noise) signal. In practice, surveyors first sample heights in an area, then use these to produce a Digital Land Surface Model in the form of a TIN. The DLSM can then be used to visualize terrain, drape remote sensing images, quantify ecological properties of a surface or extract land surface objects. The contour data or any other sampled elevation datasets are not a DLSM. A DLSM implies that elevation is available continuously at each location in the study area, i.e. that the map represents a complete surface. Digital Land Surface Models should not be confused with Digital Surface Models, which can be surfaces of the canopy, buildings and similar objects. For example, in the case of surface models produces using the lidar technology, one can have several surfaces – starting from the top of the canopy to the actual solid earth. The difference between the two surface models can then be used to derive volumetric measures (height of trees etc.).
Of increasing concern since the isolation of natural radium by Marie and Pierre Curie in 1898—and the subsequent advent of nuclear physics and nuclear technologies—are radiological poisons. These are associated with ionizing radiation, a mode of toxicity quite distinct from chemically active poisons. In mammals, chemical poisons are often passed from mother to offspring through the placenta during gestation, or through breast milk during nursing. In contrast, radiological damage can be passed from mother or father to offspring through genetic mutation, which—if not fatal in miscarriage or childhood, or a direct cause of infertility—can then be passed along again to a subsequent generation. Atmospheric radon is a natural radiological poison of increasing impact since humans moved from hunter-gatherer lifestyles and cave dwelling to increasingly enclosed structures able to contain radon in dangerous concentrations. The 2006 poisoning of Alexander Litvinenko was a notable use of radiological assassination, presumably meant to evade the normal investigation of chemical poisons. Poisons widely dispersed into the environment are known as pollution. These are often of human origin, but pollution can also include unwanted biological processes such as toxic red tide, or acute changes to the natural chemical environment attributed to invasive species, which are toxic or detrimental to the prior ecology (especially if the prior ecology was associated with human economic value or an established industry such as shellfish harvesting).

==== Gomori-positive astrocytes and brain dysfunction ==== A type of astrocyte with an aging-related pathology has been described over the last fifty years. Astrocytes of this subtype possess prominent cytoplasmic granules that are intensely stained by Gomori's chrome alum hematoxylin stain, and hence are termed Gomori-positive (GP) astrocytes. They can be found throughout the brain, but are by far the most abundant in the olfactory bulbs, medial habenula, dentate gyrus of the hippocampus, arcuate nucleus of the hypothalamus, and in the dorsal medulla, just beneath the area postrema. Gomori-positive cytoplasmic granules are derived from damaged mitochondria engulfed within lysosomes. Cytoplasmic granules contain undigested remnants of mitochondrial structures. These contents include heme-linked copper and iron atoms remaining from mitochondrial enzymes. These chemical substances account for the pseudoperoxidase activity of Gomori-positive granules that can utilized to stain for these granules. Oxidative stress is believed to be cause of damage to these astrocytes. However, the exact nature of this stress is uncertain. Brain regions enriched in Gomori-positive astrocytes also contain a sub-population of specialized astrocytes that synthesize Fatty Acid Binding Protein 7 (FABP7). Indeed, astrocytes in the hypothalamus that synthesize FABP7 have also been shown to possess Gomori-positive granules. Thus, a connection between these two glial features is apparent.
== History == The precise origin of absinthe is unclear. The medical use of wormwood dates back to ancient Egypt and is mentioned in the Ebers Papyrus from around 1550 BC. Wormwood extracts and wine-soaked wormwood leaves were used as remedies by the ancient Greeks. Moreover, some evidence exists of a wormwood-flavoured wine in ancient Greece called absinthites oinos. Lucretius' De Rerum Natura (I, 936–950) says that a drink containing wormwood was given as medicine to children in a cup with honey on the brim to make it drinkable. The first evidence of absinthe, in the sense of a distilled spirit containing green anise and fennel, dates to the 18th century. According to popular legend, it began as an all-purpose patent remedy created by Dr. Pierre Ordinaire, a French doctor living in Couvet, Switzerland, around 1792 (the exact date varies by account). Ordinaire's recipe was passed on to the Henriod sisters of Couvet, who sold it as a medicinal elixir. By other accounts, the Henriod sisters may have been making the elixir before Ordinaire's arrival. In either case, a certain Major Dubied acquired the formula from the sisters in 1797 and opened the first absinthe distillery named Dubied Père et Fils in Couvet with his son Marcellin and son-in-law Henry-Louis Pernod. In 1805, they built a second distillery in Pontarlier, France, under the company name Maison Pernod Fils. Pernod Fils remained one of the most popular brands of absinthe until the drink was banned in France in 1914.
== Protein == The expressed peptide contains a three-turn α-helix, and has no symmetry. The length of the peptide depends on where it is produced. If it is produced inside the mitochondria it will be 21 amino acids long. If it is produced outside the mitochondria, in the cytosol, it will be 24 amino acids long. Both peptides have been shown to have biological activity. The full length 24 amino acid version of humanin has the sequence MAPRGFSCLLLLTSEIDLPVKRRA, with four residues cleaved from the C terminal end to form the 20 amino acid mitochondrial version MAPRGFSCLLLLTSEIDLPV. Several other natural isoforms are also known with slight variations to the amino acid sequence. Synthetic versions often have one or more amino acid changes to increase stability and improve activity, such as HNG with a S→G substitution at position 14, and HNGF6A which has an F→A substitution at position 6 as well. Humanin-glycine (HNG) is up to 1000x more potent than the natural version depending how the activity is assayed, and so is one of the more common synthetic analogues used in research and investigated for potential clinical applications. Shorter fragments such as HN(1-19) and HN(1-17) also retain some activity but are generally less potent.

Bovine pancreatic ribonuclease, also often referred to as bovine pancreatic ribonuclease A or simply RNase A, is a pancreatic ribonuclease enzyme that cleaves single-stranded RNA. Bovine pancreatic ribonuclease is one of the classic model systems of protein science. Two Nobel Prizes in Chemistry have been awarded in recognition of work on bovine pancreatic ribonuclease: in 1972, the Prize was awarded to Christian Anfinsen for his work on protein folding and to Stanford Moore and William Stein for their work on the relationship between the protein's structure and its chemical mechanism; in 1984, the Prize was awarded to Robert Bruce Merrifield for development of chemical synthesis of proteins.
The 2-aza-β-tyrosine subunit of kedarcidin chromophore is altogether unknown in any other natural product; this lack of precedence frustrates any attempt at a priori identification of the genes responsible for synthesizing this structure. However, six genes are conserved among the biosynthetic clusters of kedarcidin, C-1027, and maduropeptin—while these later two enediynes do not contain a 2-aza-β-tyrosine subunit, they do feature similar (L)-tyrosine-derived components, leading Shen et al. to propose a pathway for the synthesis of the corresponding kedarcidin subunit beginning with 2-aza-L-tyrosine. This α-amino acid is thus believed to be converted to the corresponding β-amino acid by KedY4, an aminomutase encoded in the ked cluster. The resulting product is believed to be loaded onto the peptidyl carrier protein KedY2 and subsequently chlorinated by KedY3, an flavin adenine dinucleotide-dependent halogenase.
=== Financial markets === While futures for the Dow Jones Industrial Average, the Nasdaq-100, and the S&P 500 fell before U.S. stock exchanges opened on the day after Powell's announcement, all three indexes ultimately closed higher at the end of trading on January 12. However, the U.S. Dollar Index showed the dollar falling against other major currencies in the foreign exchange markets, while the price of gold in commodity markets hit a record high, and the yield on the 10-year United States Treasury note rose, and the yield curve between the 2-year and 10-year Treasury notes steepened. The Dow Jones, the Nasdaq, and the S&P 500 saw sell-offs over the next two days with concerns among investors about Federal Reserve independence contributing to the declines, along with news about the Trump administration's response to the 2025–2026 Iranian protests, comments by Trump that Microsoft would make changes to absorb the electricity costs of the data centers used for its artificial intelligence products, as well as Trump's proposed ceiling on credit card interest rates and continued calls for the United States to acquire Greenland. The dollar weakened further on January 14 after recovering the day before on the release of U.S. Consumer Price Index data, both gold and silver prices hit record highs on both days, while yields on 2-year and 10-year Treasury notes fell despite break-even inflation on the 10-year Treasury note rising on January 13.
== Early life and education == Wayne Woolley was born in Raymond, Alberta, the son of Americans living in Canada. His extended family were prominent members of the Church of Jesus Christ of Latter-day Saints; his great-grandfather was Edwin Dilworth Woolley, was a prominent Latter-day Saint bishop in Salt Lake City. Wayne Woolley (as he was known) was a precocious child who finished high school at age 13, and completed an undergraduate degree in chemistry at the University of Alberta at age 19. He pursued graduate studies in the department of agricultural chemistry at the University of Wisconsin, where he earned his PhD in 1939. His graduate research with Conrad Elvehjem concerned nicotinic acid as a treatment for canine blacktongue, with implications for human pellagra.

It targets the growth hormone-releasing hormone receptor on pituitary somatotroph cells. Binding stimulates cyclic AMP signaling and growth hormone secretion. This is the same receptor used by endogenous GHRH.
It does not act directly on adipose tissue as a primary mechanism. Instead, it increases endogenous growth hormone, which then influences lipolysis and fat distribution. The reduction in visceral fat is an indirect pharmacodynamic effect.
Tesamorelin acts upstream at the pituitary to amplify natural pulsatile growth hormone release. Growth hormone injections provide exogenous hormone and bypass pituitary regulation. The two approaches therefore differ in feedback control and hormonal dynamics.
tesamorelin 属 GHRH 类似物,作用于垂体受体以促进内源生长激素释放;生长激素本身是直接补充的外源激素。两者在给药逻辑、作用位点和反馈调控路径上并不相同。
